Effect of Oryeongsan on Renal Dysfunction in ischemia/reperfusion-induced acute Renal Failure Rats

|
Renal ischemia-reperfusion injury(IRI), an important cause of acute renal failure(ARF), increases renal tubular injury. Oryeongsan(OS), the oriental chinese herbal formula, widely used for the treatment of nephrosis, dropsy and uremia. This study investigated the effect of OS on rat model of ischemic ARF. Ischemia was induced by clamping the two renal arteries for 45 min followed by reperfusion. Peritoneal injection of 10 and 50 mg/kg/day OS for ischemia-reperfusion injury rat and IRI injury was measured 4 days after OS treatment respectively. Treatment with OS significantly ameliorates urine volume, urine osmolality, creatinine clearance(Ccr) and blood urea nitrogen(BUN) of kidney index. In addition, OS increases the expression aquaporine 2(AQP 2), Na+-K+ATPase and urea transporter(UT-B). Histopathologic studies have shown that OS improves glomerular expansion and tubular damage in the kidney cortex and medulla. These findings suggest that OS ameliorates tubular injury including tubular dysfunction in IRI induced ARF rats.
